Background: The primary objective of radiation therapy (RT) is the destroying of most tumor cells with the minimal damage of surrounding tissues. Methods of RT for different tumor types are developed according to experimental studies and clinical trials. Our purpose was to study the morphofunctional characteristics of sarcoma M-l at tumor growth different stages at control rats and after single y-irradiation with 30 Gy. Methods: The research was carried out on rats with sarcoma M-l implanted under the thigh skin. On the 16th day after implantation tumors were exposed to local single y-irradiation by 60Co at dose rate 0.8 Gy/min. Dissection was done after 3 hours, and 1, 3 and 10 days after irradiation. The techniques included immunostaining of PCNA, detection of the nucleolar organizers regions (AgNORs) and apoptotic cells on samples impregnated by Moser. Morphometric investigations were carried out on microscopic image analysis system. Results: The exposure led to partial regression and subsequent fast re-growth of tumors, with full recovery of volumes in 10 days after irradiation. By the 3rd day after impact the numerical density of viable tumor cells in test area decreased 2-fold. While their proliferative activity remained very high. AgNORs activation during this period indicates that biosynthetic processes are directed to the repair of survived tumor cells damaged structures. Quantitative analysis showed that in peripheral zone of tumors, proliferating cells fraction by PCNA index was 76.5%, and level of spontaneous cell death by apoptosis index was 0.28%. Individual variations of irradiation effect on sarcoma M-l are due to tumor volume at the impact time. During the post-radiation growth of sarcoma PCNA index decreased by 18.6%, while apoptotic index increased by 5-fold. The quantification of markers expression used in oncoradiobiology, improves the objectivity of obtained data, and can be applied to develop radiation and combined therapy regimens in experimental models of tumor growth.
